骨保护素(osteoprotegerin,OPG)和核因子NF—κB受体激活剂(receptor activator of nuclear factor—κB,RANK)及其配体(receptor activator of nuclear factor—κB ligand,RANKL)是近年来发现的与骨吸收和重建密切相关的细胞因子,在关节炎骨破坏中起重要作用。RANKL和RANK结合可促进破骨细胞(osteoclast,OC)及其前体细胞分化成熟和活化;而OPG可阻断此过程而抑制RANKL介导的OC分化,从而减少骨质损害。
研究目的:本课题以骨关节炎(osteoarthritis,OA)和健康者为对照,通过观察类风湿关节炎(rheumatoid arthritis,RA)和强直性脊柱炎(ankylosing spondylitis,AS)患者滑膜、滑液和外周血中RANK、RANKL和OPG的表达、分布及与骨破坏的关系。进一步阐明RANK、RANKL和OPG系统在RA和AS关节骨侵蚀发生机制中的中心作用,为临床早发现、早评价骨破坏程度,早使用OPG预防骨破坏提供理论依据。

Osteoprotegerin(OPG), receptor activator of nuclear factor—κB (RANK) and receptor activator of nuclear factor—κB ligand(RANKL) are the cytokines which have close relationship with the absorption and re-establishment of bones.
