红阳猕猴桃(A.chinensis var rufopulpa Liang et Ferguson)是在四川猕猴桃原产地苍溪发现的自然变异品种,具有极高的推广前景,其红肉的性状极具开发利用价值。为加快这一珍贵资源的繁殖,本研究以红阳猕猴桃为试材,通过田间植株顶芽、带芽茎段、茎段、叶片、叶柄以及试管苗叶片等多种外植体离体培养,研究其诱导成苗、幼苗增殖、生根移栽技术,以建立红阳猕猴桃快速繁殖体系。结果如下:
(1)通过诱导丛生芽再生成苗的途径效果差被淘汰,而选择通过愈伤组织诱导成苗途径。
(2)愈伤组织成苗途径的适宜外植体为去皮茎段,取材最佳季节是夏季。不同外植体产生愈伤组织、芽分化率差异大,茎段愈伤组织分化率最高达83.73%;叶柄的愈伤组织芽再生率12.88%;叶片的愈伤组织芽再生率仅为8.10%。
(3)带皮茎段离体培养,筛选出最佳培养基是:
