目的:研究静磁场(static magnetic fields,SMF)联合顺铂对K562细胞杀伤作用的机制;应用高效液相色谱(HPLC)检测磁场处理前后K562细胞对顺铂的摄取量,比较细胞对顺铂的摄取量与细胞活力间的关系;通过彗星电泳检测,分析静磁场联合顺铂致人外周血淋巴细胞DNA损伤情况,比较其与K562细胞的异同。
方法:
1.MTT法检测磁场联合不同浓度顺铂处理K562细胞(人红白血病细胞)12h内细胞活性变化趋势;MTT法检测12h内磁场联合顺铂处理K562细胞活性随时间变化的趋势。
2.HPLC法检测了12h内K562细胞对顺铂的摄取;彗星电泳检测了12h内SMF联合顺铂对K562细胞DNA的损伤;并用彗星电泳检测了SMF联合顺铂处理人静息淋巴细胞12h细胞DNA损伤情况。
结果:
