目的建立自身免疫性肝炎小鼠模型,研究鼠纤维介素(murine fibrinogen like protein 2, mfgl2)在实验性自身免疫性肝炎(experimental autoimmune hepatitis, EAH)的表达以及意义,并进一步研究其在发病机制中的作用。
方法采用易感动物C57BL/6小鼠,在第1天和第7天分别以新鲜制备的含蛋白质浓度为0.5-2g/L的S-100肝抗原0.5ml与等体积的完全弗氏佐剂(complete Freund's adjuvant, CFA)充分乳化后予腹腔注射,建立了自身免疫性肝炎小鼠动物模型。分别于造模后7d、14d、21d、28d、35d取小鼠外周血和肝脾组织,进行常规病理组织学检查,免疫荧光法检测血清中的自身抗体。免疫组织化学、逆转录多聚酶链反应(reverse transcription polymerase chain reaction, RT-PCR)和流式细胞仪方法测定脾脏组织中mfgl2的表达。
