On The Role and Compatibility Effect of Yuzhang No.Ⅱ Recipe to Aplastic Anemia Model Mouse Based on The PTEN/AKT/mTOR Signaling Pathway of Marrow Cells
目的:通过研究愈障Ⅱ号方及其拆方配伍对再障模型小鼠骨髓造血细胞PTEN/AKT/m TOR信号转导通路的影响,探讨愈障Ⅱ号方治疗再障的作用机制及拆方配伍意义。方法:SPF级BALB/c雌性小鼠75只,随机选取15只为空白组,其余采用乙酰苯肼、X射线及环磷酰胺复制再障小鼠模型,再障造模成功后按体质量随机分为模型组、全方组、补肾组及调和肝脾组,并采用束缚制动加控制饮食法建立肝郁脾虚模型,同时全方组、补肾组及调和肝脾组小鼠给予中药灌胃,模型组小鼠给予等量生理盐水灌胃。实验期间观察小鼠一般状态,检测小鼠外周血细胞计数。灌胃16d后取材,观察血细胞、骨髓细胞形态,行骨髓病理;流式细胞术检测骨髓单个核细胞中CD45~+CD117~+造血干细胞比例及骨髓单个核细胞凋亡率;RT-PCR法检测骨髓单个核细胞中PTEN、AKT、m TOR m RNA表达;Western blot法检测骨髓单个核细胞中PTEN、p-PTEN、AKT、p-AKT、m TOR、p-m TOR、p-P70S6K、p-4E-BP1表达。结果:1.愈障Ⅱ号方能显著改善再障小鼠的一般状态,全方组明显优于拆方组(P<0.01)。补肾组对再障小鼠行为状态、大便状...
