目前乙型肝炎病毒感染是全球性公共卫生问题,现有3.5亿患者受其困扰。随着抗HBsAg单克隆抗体和抗体片段的研究开发,发现能够和乙肝病毒表面特异性位点结合并封闭其侵袭肝细胞的活性,而融合了对病毒有较好临床治疗效果的干扰素γ的双功能抗体将能够起到导向性预防和治疗的双重作用。本文在前期工作的基础上,将抗HBsAg单链抗体(single-chain Fv against HBsAg,HBscFv)基因与干扰素γ(IFNγ)基因在体外拼接成单一基因,然后转入巴氏毕赤酵母中分泌表达,并对表达产物进行了纯化及体外生物学活性和理化性质鉴定。
体外基因及其表达载体的构建。首先是设计合适的引物并分别从含干扰素γ基因的载体和含抗HBsAg单链抗体的质粒中PCR扩增出干扰素γ基因和HBscFv基因。接着成功采用重叠PCR技术,通过基因SOEing方法将两基因拼接成单一基因HBscFv-IFNγ,并进行序列测定,其结果表明,所获得的基因产物为正确的HBscFv-IFNγ基因。将此HBscFv-IFNγ基因插入真核高效表达的载体pPICZaA中,获得含目的基因的表达载体pPICZaA/HBscFv-IFNγ,并...

Hepatitis B virus infection is a worldwide public health problem affecting about 350 million people per year. Monoclonal antibodies or their fragments are only useful in prevention if they bind to and block surface regions of the virus.
