新城疫(NDV) La Sota和V4疫苗是新城疫防制中最为常用的两种疫苗,但是实践应用和研究中发现,尽管在诱导产生体液免疫应答方面La Sota优于V4疫苗,V4疫苗却能产生与La Sota疫苗相似的免疫效果。是什么机制导致这种现象的出现,一直困扰着人们。为从根本上阐明其中的机制,优化现有疫苗,更好的防制ND,我们在体液免疫和粘膜免疫研究的基础上,进一步应用流式细胞技术对NDV La Sota与V4株免疫鸡及免疫攻毒鸡CD4~-T、CD8~-T细胞亚群动态变化进行了比较。结果表明,总体上V4疫苗免疫后外周血和脾脏中CD4~-T细胞、CD8~-T细胞百分含量高于La Sota组,提示V4株诱导的T细胞免疫应答要优于La Sota株,进而从细胞免疫方面揭示了两种疫苗激发的免疫应答的差异。试验还表明新城疫La Sota、V4疫苗免疫后1天,外周血中的CD4~-T细胞明显增加,即激发了细胞介导的免疫反应,因此在疫苗免疫后抗体产生前,细胞免疫在对强毒攻击中可能发挥着重要的作用。
