天然药物五倍子在大鼠龋病模型中的防龋作用

【摘要】 目的:利用大鼠龋病模型,对前期实验获得的五倍子防龋有效成分—五倍子E(总鞣质)和五倍子B(没食子酸和没食子酸甲酯)进行动物实验,为五倍子进入临床试验提供实验依据。方法:(1)建立大鼠龋病模型:选择17天断奶SPF-SD大鼠,喂以氨苄西林钠抑制其内源性细菌,以远缘链球菌6715标准株接种于大鼠口腔,饲以致龋食谱Keye2000#,5周后可见大鼠磨牙出现龋损。(2)实验分组:实验组为:五倍子E(8000ppm)、五倍子B(4000ppm)和五倍子B(4000ppm)+氟化钠(250ppm);阴性对照组为蒸馏水,阳性对照组为氟化钠(250ppm)和氯已定(0.12%)。(3)实验过程:按照实验分组,于大鼠接种远缘链球菌第27天以不同药物处理口腔,每天2次,连续5周。实验结束时采集大鼠刺激性唾液,制备牙颌骨标本,记录唾液远缘链球菌水平和 Keyes龋齿记分。实验过程中同时记录安全性评价指标。结果:(1)远缘链球菌水平:各实验组远缘链球菌水平较阴性对照组有不同程度降低(P<0.05),组间无统计学差异(P>0.05)。(2)龋齿记分:各实验组药物处理对大鼠龋齿形成及严重程度均产生不同程度抑制作用(P<0.05),但均低于氟化钠组。其中五倍子E组、五倍子B组、五倍子B+氟化钠、氟化钠组对光滑面龋抑制程度分别为49%,43%,50%,62%;对窝沟龋抑制程度分别为35%,17%,19%,41%。五倍子B+氟化钠组未见明显协同作用,两者单独应用无统计学差异(P>0.05)。(3)初步安全性评价:通过对体重、进食情况、口腔黏膜刺激症状等指标的观察,表明实验药物经口腔局部应用,在实验周期内大鼠健康状况良好,未见明显毒副反应。结论:五倍子E和B在大鼠口腔局部使用,具有较好的防龋效果,实验期内未见明显毒副反应,为五倍子的临床应用奠定了实验基础。

【Abstract】 OBJECTIVE: The aim of this study was to evaluate the anticaries potency of Galla chinesis (GCE) on dental caries development and plaque microbiology in rats. METHODS: The caries models in rats were established as follows: The SPF-SD rats were infected with S.sobrinus 6715 when 22,23 and 24 days after birth, and were fed with diet 2000 and 5% sucrose in drinking water ad libitum until the end of this experiment. Treatment groups were GCE (8000ppm), GCE-B (4000ppm) and GCE-B (4000ppm) + fluoride (250ppm). Negative control was distilled water, while positive control was fluoride (250ppm) and chlorhexidine (CHX) (0.12%). Topically treatments were done for every group twice daily. The level of S. sobrinus in saliva and Keyes scores representing the incidence of caries were recorded at the end of the experiment. RESULTS: The level of S.sobrinus in CHX group showed statistical significance when compared with the negative control (P<0.05). GCE (49% reduction in smooth-surface caries while 35% reduction in sulcal-surface caries) and GCE-B (43% reduction in smooth-surface caries while 17% reduction in sulcal-surface caries) reduced caries formation when compared with negative control (P<0.05). The toxicity of GCE and GCE-B were tested by observation of food and water consumption, weight gained, general and oral mucous appearance, it was proved that GCE and GCE-B showed no obvious toxicity during the experiment when used topically in rat mouth. CONCLUSIONS: Gaila Chinesis could reduce S.sobrinus inoculation into the rat mouths, and had significant anticaries potency in rat.
