玻璃化保存液对黏附成骨细胞形态及成活率的影响

【摘要】 利用自行设计的细胞渗透性实验台,实时观察了黏附成骨细胞在高浓度玻璃化保护液中的形态变化。经过七步添加和七步去除95%的VEG玻璃化保护液后,黏附的成骨细胞的形态由细长的纺锤形变为粗短的圆柱形,说明黏附细胞的两端及边缘的黏附点已脱落,但经过3-4小时的培养后,黏附细胞逐渐恢复了原来的形态,表明玻璃化保护液的处理只是改变了细胞的形态,没有造成黏附细胞的脱落和严重损伤。
【Abstract】 The morphology changes of attached osteoblast cells exposed to vitrification solutions of various concentrations were observed by use of a diffusion stage.After addition and removal of 95%VEG by a seven step stepwise method, the morphology of attached cells changed from a long podia or spindle-like appearance to short cylinder shape.The cells appeared to lose contact with the scaffold at the periphery of the cell, perhaps releasing focal adhesions and becoming more centralized.When incubated for 3-4 hours, the attached cells retained their original morphology .The results indicated that vitrification solution treatment changed only the morphology of attached cells, but did not damage the cells or cause detachment of cells.
