半胱氨酸自组膜免疫传感器的研究

【摘要】 在金片电极上制备半胱氨酸自组装单分子膜(SAM),利用戊二醛交联技术将抗体固定在自组膜修饰电极上,并利用循环伏安法分别对裸金电极、自组膜修饰电极、抗体膜电极以及抗原抗体反应后的电极进行了表征,最后根据抗原、抗体反应前后电极电位的变化,对游离抗原进行检测,电极检测下限可达0.05ng/ml,检测范围为0.05.5.4ng/ml。
【Abstract】 A procedure for the covalent coupling of antibody (anti-IgG) to a gold surface modified with Self-assembled monolayer (SAM) of cysteine was investigated. Each step in the modified gold electrode was followed by an electrochemical characterization; the cyclic voltammograms were obtained with the same bare or modified Au electrode as the working electrode. The immunosensor could detect the IgG in a range of 0.05-5.4ng/mL within 13 min. In a word, the self-assembled monolayer using cysteine, which was cheaper, was a better alternative for immunosensor applications when SAMs were used.
