益气养阴、活血化瘀方对病毒性心肌病小鼠心肌MMP-1 mRNA表达的影响

【摘要】 目的观察益气养阴、活血化瘀方对病毒性心肌病小鼠心肌基质金属蛋白酶1(matrix metalloproteinase-1,MMP-1)mRNA表达的影响。方法采用反复腹腔注射柯萨奇B3m病毒的方式复制动物模型;270只Balb/c小鼠随机分为5组,空白组30只,模型组、中药高剂量组、中药低剂量组、西药组各60只;HE和VG染色观察小鼠心肌形态学变化;RT-PCR法检测心肌组织MMP-1 mRNA的表达水平。结果益气养阴、活血化瘀方能明显改善病毒性心肌病小鼠心肌病理形态及超微结构的变化,下调心肌组织MMP-1 mRNA的表达水平,以中药高剂量组效果最显著,MMP-1为0.35±0.11。结论益气养阴、活血化瘀方能通过下调MMP-1 mRNA的表达,抑制细胞外基质降解及心肌重构,这可能是其防治病毒性心肌病的作用机制之一。

【Abstract】 Objective To observe MMP1mRNA expression in the myocardial tissue of mice with viral myocardial disease(VMD)treated with compound prescription of replenishing qi and nourishing yin, invigorating blood circulation and removing blood stasis. Methods The animal models of VMD were cstablished by injecting CVB3m to the abdominal cavity of Balb/c mice repetitively. 270 Balb/c mice were divided into five groups randomly.The morphological changes of myocardium were detected MMP-1 mRNA expression in myocardial tissue were observed with RT-PCR method. Results The compound prescription could improve the of pathomorphology and ultrastructure of myocardium in VMD mice, lower MMP1mRNA expression in the myocardial tissue of VMD mice. The effects were better in high dose treated group . Conclusion The compound prescription could lower MMP-1 mRNA expression in the myocardial tissue of VMD mice, inhibit the degradation of extracellular matrix and myocardium remodeling. This might be one of the way of preventing and treating VMD.
