三七总皂苷对人脐静脉内皮细胞的促血管新生作用
Proangiogeneic effect of total saponns of Panax notoginseng in human umbilical vein endothelial cells (HUVEC)
【摘要】 目的:研究三七总皂苷(PNS)对人脐静脉内皮细胞(HUVEC)的增殖作用。方法:采用体外培养HUVEC的方法,通过XTT法以及细胞计数法观察对细胞的增殖作用;细胞侵入实验观察细胞迁移的能力。结果:PNS对HUVEC有促增殖作用,XTT实验中100μg/ml浓度增殖率为(33.58±2.41)%;在细胞计数实验100μg/ml浓度增殖率为(94.15±10.96)%;在细胞侵入实验中100μg/ml浓度侵入率为(42.64±9.50)%。结论:PNS能够促进HUVEC的增殖与侵入。
【Abstract】 Objective: To investigate the proliferation efficiency of total saponins of Panax notoginseng (PNS) on human umbilical vein endothelial cell (HUVEC). Methods: HUVEC were incubated in vitro. The extract was identified to stimulate the proliferation of HUVEC by XTT assay and microscopic cell counting; and the numbers of invaded cells and the mean tube length were significantly increased in both treatment groups; Results: The HUVEC cells increased proliferation in the response to stimulation of PNS. In the XTT assay, after being incubated with PNS 100μg/ml, the increasing rate was(33.58±2.41)%; In the cell counting assay, the cells were raised by(94.15±10.96)% at the concentration of PNS 100μg/ml; And in the invasion assay, the PNS could induce the cells’ invasion with the concentration of 0.10μg/ml, 10μg/ml, 100μg/ml, and there was(42.64±9.50)% enhanced invasion at the concentration of 100μg/ml. Conclusion: PNS can promote the proliferation and invasion of HUVEC cells.
