逍遥散抗抑郁作用研究
Studies on the anti-depression effect of Xiaoyao Powder(逍遥散) in mice
【摘要】 目的:探讨逍遥散的抗抑郁作用及机制,为其疏肝解郁功效及临床应用提供药理学参考。方法:采用小鼠悬尾实验和小鼠强迫游泳实验观察逍遥散的抗抑郁作用,采用开场实验观察逍遥散对小鼠自主活动的影响;采用5-HTP、DOI致小鼠甩头实验探讨逍遥散抗抑郁作用的机制。结果:逍遥散7.8、15.6g/kg剂量连续给药7d能明显缩短悬尾实验中小鼠的不动时间,15.6g/kg剂量连续给药7d能明显缩短强迫游泳实验中小鼠的不动时间,各有效剂量对小鼠自主活动无影响;逍遥散31.2g/kg剂量连续给药7天能显著减少5-HTP所致小鼠甩头次数,但对DOI诱导的小鼠甩头反应无明显影响;逍遥散31.2g/kg剂量单次给药能明显减少由5-HTP、DOI诱导的小鼠甩头次数;逍遥散15.6g/kg剂量单次给药亦能显著减少由5-HTP诱导的小鼠甩头次数。结论:逍遥散具有抗抑郁作用,作用机制可能与影响脑内5-HT能神经系统有关,特别是5-HT2A受体的功能。
【Abstract】 Objective: To investigate the anti-depression effect and the possible mechanism of Xiaoyao Powder in several animal models. Method:The tail suspension test (TST) and the forced swimming test (FST) were applied to study the anti-depression effect of Xiaoyao Powder in mice. The open-field test was carried out to observe the effect of Xiaoyao Powder on automatic activities in mice. The possible mechanism of its anti-depression effect was studied by 5-HTP or DOI induced head-twitch-response (HTR) in mice. Result: The immobility time of TST was significantly decreased by the dose of 7.8、15.6g/kg of Xiaoyao Powder (P<0.01), as well as the immobility time of FST was significantly decreased by the dose of 15.6g/kg of Xiaoyao Powder(P<0.01) following treatment for 7d continuously in mice. Xiaoyao Powder had no effect on the automatic activities in the open field test in mice at the dose of 7.8,15.6g/kg. The times of 5-HTP induced HTR were significantly decreased by the dose of 31.2g/kg of Xiaoyao Powder following treatment for 7d continuously in mice(P<0.05), but had no effect on DOI induced HTR. The times of 5-HTP and DOI induced HTR were significantly decreased by the dose of 31.2g/kg of Xiaoyao Powder single treatment in mice (P<0.01, P<0.05) , as well as the times of 5-HTP induced HTR were also significantly decreased by the dose of 15.6g/kg of Xiaoyao Powder single treatment in mice (P<0.01). Concluslono:The results suggest that Xiaoyao Powder has the anti-depression effect, which the mechanism could be related to affect the 5-HT nervous system, especially the 5-HT2A receptor.
