经胎盘接种小鼠巨细胞病毒对仔鼠听力的影响研究
The researchment of murine inborn hearing change after microinjecting cytomegalovirus via the placenta way.
【摘要】 目的观察胎盘接种建立小鼠巨细胞病毒对仔鼠的听力的影响。方法MCMV Sm ith标准株经小鼠胚肺细胞增殖传毒后,用ELISA筛选无MCMV感染史性成熟期小鼠,雌雄同笼受孕。孕龄12.5天时,模型组胎盘显微注射MCMV,手术对照组注射DMEM培养基,空白对照组只手术开腹,清点孕囊,不做任何特殊处理。待母鼠自然分娩后,随机选取4周龄仔鼠,应用听性脑干反应(aud itory brainstern response,ABR)检测听力水平。结果模型组与其他各组比较,ABR I波阈值明显上升,差异有显著意义(F=5.984,P=0.009)。结论经胎盘接种MCMV可以引起仔鼠的听力损伤。
【Abstract】 Objective:Observe the murine inborn hearing change through the microinjection way.Methods: MCMV Smith strains were prepared from murine embryo fibroblast(MEF) of MCMV infected mice.The mature mice without MCMV infection identified by Enzyme-linked immunoabsorbent assay(ELISA) were served as the subject.We put the male and female together,and injected the MCMV into the placentas of the model group female mice at the day 12.5 of gestation,the operation control group were injected with DMEM medium,the blank control group were only counted the conceptus by surgery.Randomly choosing offsprings at 4 week′s old and detecting their auditory function with auditory brainstern response(ABR).Results:The modle group′ response threshold of ABR I was much higher than that of other two groups.There was significant difference to other two groups(F=5.984,P=0′009).Conclusion: MCMV-microinjection via the placenta can cause inborn hearing loss.
