【摘要】 目的探讨不同浓度苦参碱对体外培养的人胃癌细胞NCI-N87的增殖抑制作用。方法NCI-N87细胞经不同浓度苦参碱处理后,采用CCK-8法检测药物对肿瘤细胞的抑制作用,观察细胞形态学改变,检测细胞DNA分布。结果苦参碱浓度>3.0mg/ml,细胞毒作用较大;浓度在0.5～2.0mg/ml时,对肿瘤细胞的生长有抑制作用,且呈时间剂量依赖性;浓度在0.1mg/ml时,对肿瘤细胞生长几乎无抑制作用。苦参碱作用后可增加G1期细胞所占的百分比,阻止细胞进入S期。结论苦参碱对人胃癌细胞NCI-N87增殖有抑制作用,并能阻抑细胞周期的进程。更多还原

【Abstract】 Objective:To investigate the inhibition of different concentrations of matrine on Proliferation of human gastric cancer cell line NCI-N87 cultured in vitro.Methods:NCI-N87 cells were treated by matrine in different concentrations and variablc durations.Cell growth and proliferation were observed by CCK-8 assay.Morphological changs of cell were observed,and cell-DNA distribution was assayed.Results:When the drug concentration was higher than 3.0mg/ml,the major effect was cell mecrosis.Matrine in the concentrations from 0.5mg/ml to 2.0mg/ml could inhibit proliferation of NCI-N87 cells as reveled by CCK-8 assay.Increase in the percentage of Gl phase cell was observed by flow cytometry.Conclusion:Matrine can inhibit the proliferation of human gastric cancer cell line NCI-N87 efficiently by affecting the cycle of cells.更多还原
