氧化苦参碱对兔油酸型急性肺损伤的保护作用研究

【摘要】 目的:研究氧化苦参碱对急性肺损伤的保护作用并探讨其机制。方法:采用静脉注射油酸的方法复制家兔急性肺损伤模型。动物随机分为6组:正常组、模型组、地塞米松组和氧化苦参碱组(20,40,80 mg.kg-1)。实验期间记录动物呼吸频率,不同时间点行血气分析,并与注射油酸后3 h时收集支气管肺泡灌洗液(BALF)测定白细胞介素-8(IL-8)和细胞间黏附分子-1(ICAM-1),实验结束测定肺干、湿重,计算湿/干重比,并行肺组织的病理学检查。结果:氧化苦参碱可明显减轻油酸模型组兔肺组织病理学改变,降低肺湿/干重比,升高PaO2/F iO2的比值,降低BALF中IL-8和ICAM-1的含量。结论:OMT能有效地保护油酸致家兔急性肺损伤,可能通过抑制细胞因子的过度生成从而达到肺保护作用。

【Abstract】 Objective: To investigate the effect of oxymatrine on acute lung injury(ALI)induced by oleic acid in rabbits.Methods: The rabbits were randomly divided into 6 groups: normal group,oleic acid group,dexamethasone group,oxymatrine groups(20,40 and 80 mg·kg-1).The respiratory amplitude and rate were observed,and blood samples were taken from cervical artery for blood-gas analysis at 30,60,and 120,180 min after oleic acid injection.At the end of experiment,the concentrations of interleukin-8(IL-8)and intercellular adhesion molecule-1(ICAM-1)in bronchoalveolar lavage fluid(BALF)were measured and pathologic changes of lung tissues were observed under light microscopy.Results: Continuous infusion of oxymatrine significantly alleviated oleic acid-induced lung injury accompanied by improvement of pathological changes,reduction of wet-to-dry weight ratio,increases in the ratio of partial pressure of oxygen in artery(PaO2)and fraction of inspired oxygen(FiO2),and decreases in IL-8 and ICAM-1 level in BALF.Conclusion: Oxymatrine has a beneficial effect on acute lung injury induced by oleic acid in rabbits by inhibition of inflammatory cytokine production.
