【摘要】 目的:探讨乳黄片对A型肝性脑病大鼠大脑皮层星形胶质细胞结构蛋白-神经胶质纤维酸性蛋白(CFAP)表达的影响。方法:W istar雄性大鼠96只随机分为6组,正常对照组(A组)、模型组(B组)、乳果糖组(C组)、乳黄片低、中、高剂量组(D、E、F组)。除A组外,其余各组大鼠用硫代乙酰胺(TAA)造模,在TAA首次造模前,对A、B组大鼠灌以生理盐水1m l/100g。其余各组大鼠灌以相应药物。实验30小时后取材。免疫组化法测定大鼠皮层GFAP的表达水平。结果:B组大鼠GFAP染色阳性细胞以及平均光密度较A组明显降低,两者比较P<0.01,差异有显著性意义。D、E、F组和C组大鼠GFAP均较B组显著升高(P<0.01),且D、E、F组升高程度均较C组显著(P<0.05或P<0.01),其中以中剂量组升高最明显。结论:乳黄片有良好的防治A型肝性脑病的作用,其机制可能是通过保护肝细胞降低血氨进入血脑屏障的浓度,改善星形胶质细胞受损结构,提高GFAP蛋白表达而实现的。乳黄片抗A型肝性脑病的作用位点可能为星型胶质细胞。更多还原

【Abstract】 Objective:Study effect of Ruhuang pill on expression of astroglia structure’s glial fibriliary acidic protein(GFAP) in cortex of rat with hepatic encephalopathy(HE) of type A.Methods:Ninety six male Wistar rats weighed 200±20g were randomly divided into six groups:normal control gronp(group A);model gronp(group B);lactulose gronp(group C);Ruhuang pill group of small dose(group D);Ruhuang pill group of middle dose(gropu E);Ruhuang pill group of large dose(group F);all rats except group A were induced to be modeled by administration with thioacetamide(TAA) 250mg/kg(twice,per 24 hour).Before first modeled by administration with TAA,each rat in group A and group B were given normal saline 1ml/100g weight,and the left were given the same corresponding medicine respectively.After 30 hours of experiment,all rats were sacrificed.Immunohisytochemistries were performed to examinne the expression of GFAP.Results:Ruhuang pill could up-regulate the expressions of GFAP,increase significantly the positive numbers and means of optical density compared with those in model group A(P<0.01),and those of middle dose group increased most.Compared with group B,group A was much lower in rats’ GFAP staining cells and average optical(P<0.01).There were much more significant between the two groups.Group D,E,F and group C all increased in expression of GFAP obviously than group A.Compared with group C,group D,group E and group F all increased more obviously which the middle dose group increased most(P<0.05 or P<0.01).Conclusion:Ruhuang pill has good effect on the prevention and treatment of HE of type A.The effect can be carried out by the following ways:protect liver function,decrease the concentrations of ammonia(in blood and cerebrospinal fluid) and glutamine to enter blood-brain barrier(BBB),ameliorate the injuring structure and increase protein expressions of astroglia.The function point of Ruhuang pill in the treatment of HE may be astroglia.更多还原
