冠心病患者同型半胱氨酸与PPARα γ mRNA、蛋白表达相关性分析
Research of the relationship between serum homocysteine and PPARαγ mRNA, protein expression in coronary artery disease
【摘要】 目的探讨冠心病患者同型半胱氨酸与过氧化体增殖剂激活的受体的关系。方法采用高效液相色谱法测定287例冠心病患者和50例健康体检者(健康对照组)空腹血清Hcy的水平,根据测量结果将冠心病患者Hcy的水平分为冠心病Hcy正常、轻、中、重组,实时RT-PCR、westernblot检测各组血浆单核细胞PPARα、PPARγ mRNA和蛋白表达。结果冠心病轻、中、重组患者的PPARα、PPARγ mRNA、蛋白表达明显低于健康对照组(P<0.05)和冠心病Hcy正常组(P<0.05),并与Hcy呈明显负相关。结论冠心病患者Hcy较高而PPARα、PPARγ等的表达较低,提示高Hcy血症通过某种方式抑制PPARα、PPARγ等的表达,是其导致动脉粥样硬化的机制之一。
【Abstract】 [Objective] To study the association between homocysteine (Hcy) and peroxisome proliferators activateed receptor (PPARs) [Method] The level of Hcy of 287 coronary artery disease patients and 50 healthy adults were messured by reverse-phase high performance liquid chromatography (HPLC), divided into four groups by hcy level of coronary heart disease (CAD) patients: CAD hcy normal group, CAD hcy light range group, CAD hcy midrange group and CAD hcy serious group. The PPAR α and PPAR γ mRNA, protein expression in plasma lymphomonocyte were carried out by the real time RT-PCR and western blotting respectively. [Results] Compared with the health control group and CAD hcy range group, the levels of PPAR α and PPAR γmRNA and protein were lower (P <0.05), and had a negative relativity with hcy density obviously. [Conclusion] Hcy level of the cases with CAD is higher while the mRNA and protein expressions of PPAR α and PPAR γ were lower, which indicate higher hhcy refrains the expressions of PPAR α and PPAR γ may be one of mechanisms leading to artherosclerosis.
