【摘要】 目的探讨多西紫杉醇(Docetaxel,TXT)对鼠血管肉瘤细胞株的增殖抑制作用。方法采用Alamar Blue法测定TXT对ISOS-1细胞增殖抑制的影响,并与其它抗肿瘤药物进行比较;用流式细胞仪测定细胞周期的变化。结果TXT能显著抑制ISOS-1细胞的增殖,IC50为15.8ng/ml,明显低于其它抗肿瘤药物;TXT作用后细胞周期发生明显变化,高浓度TXT对ISOS-1细胞株有G2-M期阻滞作用,低浓度TXT主要将ISOS-1细胞阻断于G0-G1期并诱导细胞凋亡。结论TXT能强烈抑制鼠血管肉瘤细胞株ISOS-1的增殖,这种抑制作用可能通过作用G2-M期及诱导细胞凋亡来实现的。更多还原

【Abstract】 Objective To study the proliferation inhibition effect of TXT on murine angiosarcoma cell line.Methods To determinate the proliferation inhibition effect of TXT on ISOS-1 cells by Alamar Blue assay,and compare TXT with other chemodrugs.The changes of cell cycle was determinated by FCM.Results TXT can inhibit the proliferation of ISOS-1 cells obviously,the IC50 was 15.8 ng/ml,which was lower than the rates of other chemodrugs.Cell cycles changed much after TXT was applied.High concentration TXT can block ISOS-1 cell line at G2-M phase,while low concentration TXT can block ISOS1 cells at G0-G1 phase to induce apoptosis.Conclusion TXT can inhibit the proliferation of murine angiosarcoma cell line ISOS-1 strongly,it can not only act on G2-M phase,but also induce apoptosis.更多还原
