人前列腺癌细胞特异性结合短肽的筛选
Screening of peptides bound specifically to human prostate carcinoma cells
【摘要】 目的获得与人前列腺癌细胞系PC3M细胞特异结合的短肽,作为人前列腺癌靶向治疗的先导化合物。方法以PC3M细胞为靶细胞,正常人前列腺细胞为吸附细胞对噬菌体7肽库进行差减筛选,用细胞ELISA、免疫荧光细胞化学染色鉴定阳性噬菌体克隆并测序。结果经三轮筛选,从随机挑选的10个噬菌体克隆中得到8个能特异性与PC3M细胞结合,而不与正常人前列腺细胞结合的阳性克隆。其氨基酸序列有一定同源性。结论得到多个序列不同的特异性结合PC3M细胞的噬菌体克隆。本实验获得的短肽具有一定的肿瘤亲合力和特异性,为针对不同位点的靶向药物设计提供了实验依据。
【Abstract】 Objective Identified peptides bound specifically to PC3M human prostate adenocarcinoma cells for organ specific drug targeting.Methods PC3M cells were screened with seven phage display peptide libraries which incubated with nomal human prostate before to get rid of bound peptides.We identified positive peptides using cell ELISA and immunofluorescence experiments and DNA sequencing.Results After three selection rounds,eight positive clones that specifically bound to PC3M cells rather than nomal human prostate cells displayed a sequence similarity.Conclusion These results indicate that identified peptides maybe favor organ specific drug targeting because they specifically recognize prostate cancer.
