IκB-α基因转染HCC9204细胞对NF-κB和MMP-9表达的影响

【摘要】 目的观察高转移性人肝癌细胞系HCC9204转染抑制性κB基因(IκB-α)后核因子-κB(NF-κB)和基质金属蛋白酶-9(MMP-9)表达的改变。方法采用脂质体法将IκB-α基因转染HCC9204细胞,应用Western-blot及RT-PCR法检测MMP-9和NF-κB的表达;通过基底膜侵袭实验检测肿瘤细胞的浸润和转移能力。结果HCC9204细胞转染pcDNA3-IκB-α后,细胞内NF-κB蛋白表达下降,同时伴随MMP-9mRNA表达水平和细胞侵袭、转移能力的明显下降。结论NF-κB活性被抑制后引起细胞侵袭、转移能力的下降可能是由于MMP-9表达下降所致。

【Abstract】 Objective To investigate the expression changes of nuclear factor kappa B (NF-κB) and matrix metalloproteinase-9 (MMP-9) in the cultured hepatocellular carcinoma cells 9204 (HCC9204) transfected with inhibitory kappa B alpha(IκB-α)vector. Methods After pcDNA3-IκB-α vector and pcDNA3 were transfected into HCC9204 by lipofectamine method, Western-blot and RT-PCR analysis were used to detect the expressions of NF-κB and MMP-9. Migration and invasion of tumor cells were assayed by fundus membrane invaded by them. Results When pcDNA3-IκB-α was transfected into HCC9204, the expression of NF-κB was decreased at the protein level, and the expression of MMP-9 mRNA and the invision and metastasis ability of transfected cells were obviously decreased. Conclusion When the activity of NF-κB is inhibited, the ability of invasion and metastasis in HCC9204 cells decrease, which could be related to the decreased the expression of MMP-9.
