高产γ-聚谷氨酸菌株的选育与鉴定
Screening and identification of strains for high production ofγ-polyglutamic acid
【摘要】 聚谷氨酸(γ-PGA)是一种生物可降解的大分子物质,广泛应用于医药、化妆品等行业。从高温处理过的腐乳中分离到1株高产γ-PGA的菌株,通过对菌落表观形态特征和电镜照片特征以及生理生化性能的观察分析,初步鉴定为革兰氏阳性枯草芽孢杆菌。该细菌最适生长温度为35℃~38℃:培养条件为蛋白胨20g/L,葡萄糖30g/L,谷氨酸钠30g/L,NaCl 5g/L,pn7.0,温度为37℃,摇床培养250r/min。经红外光谱鉴定其结果与标准品的图谱基本一致。
【Abstract】 γ-Polyglutamic acid(γ-PGA)is a kind of biodegradable and giant molecule material which can be used widely in medicine and cos- metic.A high-productiveγ-PGA strain was screened from preserved sufu which was treated in high temperature.Through the observation and analysis of the colony morphology,TEM and physiological and biochemistry characteristics,the strain was elementarily identified as Bacillus subtilis,gram-positive.The optimum temperature of this bacterium is 35℃~38℃;The fermentation condition were:peptone 20g/L,glucose 30g/L,glutamine 30g/L,NaCl 5g/L,pH 7.0,temperature 37℃,rotated cultivation 250r/min.To confirm the product isγ-PGA,Fourier trans- form infrared spectroscopy was used and the result was consistent with the standard sample.
