黑曲霉产α-半乳糖苷酶发酵条件研究
Study on fermentation conditions for α-galactosidase production by Aspergillus niger
【摘要】 对从黄酒麦曲中筛选得到的黑曲霉wx-07产α-半乳糖苷酶的发酵条件进行了研究。结果表明,蔗糖是产酶的最适碳源,胰蛋白胨是最适氮源,产酶的最佳发酵条件为在250mL三角瓶中装入75mL培养基,接种量约为106个/750mL孢子,30℃、150r/min振荡培养96h,α-半乳糖苷酶活力达到2.40U/mL,是优化前的约3.08倍。
【Abstract】 The fermentation conditions of Aspergillus niger wx-07,which was isolated from wheat Qu for the production of α-galactosidase,were studied.The results indicated that sucrose and tryptone proved to be the best candidate as carbon source and nitrogen source for the production of α-galactosidase,respectively.The optimum fermentation conditions were: 75 ml medium contained in 250 ml Erlenmeyer flask,spore inoculum of 106/750 ml,temperature 30℃,rotation speed 150 r/min and fermentation time 96 h.Under above conditions,the maximum activity of α-galactosidase was 2.40 U/ml,which was 3.08 times high than the initial level.
