损伤神经环境中小胶质细胞促进MSCs神经营养功能的信号传导通路初探

【摘要】 目的:观察损伤神经环境下,与小胶质细胞(BV2)共育后发挥神经营养功能的骨髓基质细胞(MSCs)内的信号传导。方法:采用原代细胞培养法培养骨髓基质细胞(MSCs),以BV2和PC12细胞株分别代表小胶质细胞和神经细胞进行传代培养,应用转移筛网进行BV2与MSCs的共育,流式细胞术检测PC12凋亡,W estern b lot检测磷酸化ERK、磷酸化STAT3、总ERK、总STAT3蛋白。结果:损伤神经环境及小胶质细胞同时存在时,其上清显著提高受损PC12存活率的MSCs中STAT3的激活显著增高,同对照组相比差异显著(P<0.05)。此外,STAT3激活程度与受损PC12存活率相关性的统计学检验有显著性差异(P<0.05)。结论:神经损伤环境下小胶质细胞促进MSCs神经营养功能的过程中有STAT3相关的信号传导通路的参与。

【Abstract】 Objective:We are aiming at the signal transduction in neurotrophic MSCs stimulated with microglias in the circumstances of injured neuron.Methods:Primary cell culture was adopted for MSCs culture,BV2 and PC12 were cultured as a substitute for microglias and neurons respectively,transwells were used for the co-culture of BV2 and MSCs,Flowcytometry was used for PC12 apoptosis detection,Western blot was also used for detection of phosphorylated-ERK and phosphorylated-STAT3,total ERK and total STAT3.Results:With the injured neuron circumstance and microglias,STAT3 activation was seen in MSCs,which increased PC12 survival significantly and statistically(P<0.05) compared to the control,and the relevance analysis between STAT3 activation and PC12 survival turned out to be statistically different(P<0.05).Conclusion:Under the circumstance of injured neurons,with the presence of mciroglia,STAT3-related signal transduction pathway is involved in the neurotrophic activities of MSCs.
