三种血清尿素测定方法的比较

【摘要】 目的旨在找出三种测量尿素方法之间的异同处,以利于指导临床实验。方法在Beckman Coulter Synchron CX3上用尿素酶电极法,在Beckman Coulter Synchron CX4上用尿素酶速率法,在721分光光度计上用二乙酰一肟法,对这三种方法进行方法学比较。结果三种方法之间相关性良好。尿素酶电极法线性范围大于尿素酶速率法大于二乙酰一肟法。尿素酶电极法,尿素酶速率法精密度均明显大于二乙酰一肟法,尿素酶速率法的批内变异小于尿素酶电极法,而批间变异则大于尿素酶电极法。溶血对三种方法均有影响。结论三种方法均符合临床对尿素的检测要求,各有利弊,但尿素酶电极法准确、简便、快速、范围宽,应是未来尿素测量的发展方向。

【Abstract】 Objective To explore differences among the three methods for detection urea,clinical examination.Methods Urea enzyme electrode was used on the Beckman Coulter Synchron CX3,urease velocity method was used on the Beckman Coulter Synchron CX4,and diacetyl monoxime method was used on 721 Spectrophotometer,A methodological comparison was made among the three methods.Results The three methods showed closely relationship,and the linear range of electrode method was wider than those of urease velocity method and diacetyl monoxime method.The precision of the urease electrode method and urease velocity method were both obviously better than diacetyl monoxime method,The intra-assay CV of urease velocity method was less than that of urease electrode method.But inter-assay CV of urease velocity method was higher than that of urease electrode method.Hemolysis had influence on all the three methods.Conclusion The three methods are all available to the clinic requirement for urea detection.Each measurement has its advantages and disadvantages.Especially the electrode method,which is accurate,simple,rapid,and wide in linear range,and more suitable for automatic analysis,therefore it may be trend of urea measurement in the future.
