穹窿海马伞切断与卵巢切除大鼠脑内β淀粉样肽1-40的变化
Expression of β-amyloid1~40 in rat′s brain with fornix/hippocampal fimbria transection and ovariectomy
【摘要】 目的观察穹窿海马伞切断与卵巢切除大鼠脑内不同部位β淀粉样肽1-40(Aβ1-40)表达的变化,探讨模拟实验性Alzheimer病(AD)动物模型的方法。方法成年健康雌性Wistar大鼠28只,按2×2方差分析的方法随机分为4组:穹窿海马伞切断组(FF组)、卵巢切除组(OVX组)、卵巢切除加穹窿海马伞切断组(OF组)、对照组,每组7只。穹窿海马伞切断参照Klein等的方法进行。卵巢切除参照Singh等的方法进行。对照组除不切除卵巢和不切断穹窿海马伞外,其余步骤同模型组。免疫细胞化学染色观察皮质区、海马CA1区、杏仁复合体区、基底前脑Mey-nert核等部位Aβ1-40阳性细胞表达变化。结果经F分析,双侧卵巢切除和双侧穹窿海马伞切断存在显著的主效应和交互效应(P<0.001)。双侧穹窿海马伞切断或双侧卵巢切除主效应均可导致各脑区Aβ1-40阳性细胞表达与对照组比较显著增多(P<0.01)。穹窿海马伞切断加卵巢切除二者共同作用在海马CA1区、杏仁核复合体区、Meynert核区具有显著的交互效应,表现为OF组Aβ1-40阳性细胞表达与FF组和OVX组比较显著增多(P<0.05)。结论双侧穹窿海马伞切断与卵巢切除存在显著的交互效应,表现为大鼠多个脑内Aβ1-40表达显著增多,可以作为模拟实验性AD动物模型的方法。
【Abstract】 Objective To observe expressive variety of β-amyloid1~40 (Aβ1~40) in different sections of the rat brain with fornix/hippocampal fimbria transection and ovariectomy to set up the animal model of Alzheimer′s disease.Methods 28 healthy female Wistar rats were randomly divided into 4 groups according to 2×2 ANOVA design: ①fornix/hippocampal fimbria (FF) transection group; ②ovariectomized (OVX) group; ③OVX and FF transaction (OF) group; ④control group, 7 rats in every group. The expressive varieties of Aβ1~40 immunoreactive cell in cerebral cortex, hippocampus CA1, almond complex and Meynert nucleus were observed by immunohistochemical way.Results According to F analysis, FF and OVX groups had prominently main effect in the brain areas and reciprocation effect in hippocampus CA1, almond complex and Meynert nucleus (P<0.001) resulting in the increased expression of Aβ1~40 of FF and OVX groups than that in control group (P<0.01) and the enhanced expression of Aβ1~40 in OF group than that in FF and OVX groups (P<0.05).Conclusions FF and OVX have obvious reciprocation effect, presenting significantly increased expression of Aβ1~40, which could be served as the method for establishing Alzheimer′s disease animal model.
