补阳还五汤对脑出血大鼠脑内MMP-2表达的影响

【摘要】 目的观察脑出血大鼠脑组织中基质金属蛋白酶-2(MMP-2)表达,探讨补阳还五汤治疗脑出血的作用机制。方法采用立体定向技术制作大鼠脑出血模型,对出血后2、4、7、14、21、28 d后各组脑组织进行MMP-2免疫组织化学染色,检测脑组织各区域MMP-2的阳性微血管数。结果正常组未见表达,假手术组皮质偶见MMP-2表达;模型组大鼠MMP-2呈双峰表达,在造模后4 d达最高峰(P<0.01),之后有所下降,至21 d仍有表达并再有小高峰出现(P<0.01);补阳还五汤组4 d时为表达低谷,并明显低于模型组(P<0.01),其后逐渐上升至高水平维持达2 w,其中7、14、21 d表达均高于模型组(P<0.05),28 d表达稍下降但仍高于模型组(P<0.01)。结论补阳还五汤可通过调节脑出血后MMP-2的表达,有利于微血管系统重建,改善出血后脑组织的微环境。

【Abstract】 Objective To investigate the mechanisms of Buyang Huanwu Decoction(BYHUD) by observing its effects on expressions of matrix metalloproteinase-2(MMP-2)in intracerebral hemorrhagic(ICH) rat brains.Methods ICH model was induced by injecting collagenase type Ⅶ stereotaxically into right globus pallidus.Sprague-Dawley rats were randomly divided into normal control group,sham operation group,model group and BYHUD-treated group.MMP-2 and MMP-2 positive microvessel count were detected on the 2nd,4th,7th,14th,21th and 28th day after operation.The expressions of MMP-2 were evaluated by immunohistochemistry.Results Expression of MMP-2 was not observed in normal control group and observed occasionally in the cortex of sham operation group.In model group,the expression of MMP-2 was double hump and peaked on the 4th day(P<0.01),then gradually decreased but peaked at 21th day(P<0.01) again.In BYHUD-treated group,expression of MMP-2 was lower than that in model group on the 4th day(P<0.01),then increased and was higher than that in the model group during the 7th~21th day(P<0.05).It depressed on 28th day,but was still higher than that in the model group(P<0.01).Conclusions BYHUD could regulate the expressions of MMP-2 in ICH rat brains,which might improve the microenvironment and promote the microvessel networks reconstruction after ICH.
