压力超负荷致左室肥厚大鼠心肌细胞凋亡的动态变化与腺嘌呤核苷酸转运体-1的表达

【摘要】 目的探讨心肌线粒体腺嘌呤核苷酸转运体-1(ANT1)对压力超负荷致左心室肥厚中心肌细胞凋亡的影响。方法利用腹主动脉缩窄法建立大鼠压力超负荷模型,观察时间点为术后1,2,4,7,14,21,30d。ANT1 mRNA含量的测定采用RT-PCR法,利用TUNEL法检测心肌细胞的凋亡。结果(1)与对照组比较,缩窄后4dANT1 mRNA的含量上调,并于第7天达到峰值,术后14d其含量回落至对照组水平,并保持至30d。(2)手术组心肌细胞凋亡在术后1d即升高,在4d时进入高峰期并持续至7d,其后低水平持续存在,直至实验结束。而对照组未发现凋亡的存在。结论(1)心肌细胞凋亡是心肌适应后负荷增加的一个重要的参与机制;(2)ANT1部分介导了压力超负荷致左室肥厚中心肌细胞的凋亡。

【Abstract】 Objective To explore the effects of adenine nucleotide translocator-1 (ANT1) on cardiomyocyte apoptosis in pressure overload-induced left ventricular hypertrophy. Methods The pressure overload rat model was established by abdominal aorta coarctation. The observation time points were 1, 2, 4, 7, 14, 21 and 30 days after operation. The ANT1 mRNA expression was evaluated by RT-PCR. The TUNEL method was applied to detect the cardiomyocyte apoptosis. Results 1. Compared with control group, the expression of ANT1 mRNA was upregulated on 4th day after coarctation, and peaked on 7th day, then it returned to control level on 14th day and persisted until 30th day. 2. The apoptosis increased significantly 1 day after operation, and reached a plateau between 4 and 7 day. Afterwards, it decreased continuously to a low level. However, the apoptosis was so rare that it was undetectable in control group. Conclusion Apoptosis is an important regulatory mechanism involved in cardiac adaptive response to pressure overload, and it may be mediated, in part, by ANT1.
