银杏内酯和神经生长因子对体外培养人胚肝细胞功能的影响
The effects of ginkgolide and NGF on the function of the cultured human embryonic hepatocytes in vitro
【摘要】 目的:探讨银杏内酯和神经生长因子(NGF)对体外培养人胚肝细胞功能的影响。方法:用改良的非灌流法分离人胚肝细胞,用DMEM培养基培养,分为银杏内酯组、NGF组和对照组。培养至第9d时进行糖原PAS染色,计数糖原染色阳性细胞,图象分析染色灰度;检测培养液中白蛋白和尿素的含量。STATA7.0统计软件进行方差分析和两两比较。结果:糖原染色阳性细胞银杏内酯组74.63±12.76、NGF组83.25±11.18、对照组38.25±9.44;染色灰度银杏内酯组123.19±18.96、NGF组107.25±17.09、对照组164.91±21.65;白蛋白含量(g/L)银杏内酯组3.213±0.422、NGF组3.450±0.481、对照组2.575±0.498;尿素含量(mmol/L)银杏内酯组1.813±0.290、NGF组2.013±0.344、对照组1.363±0.220。统计显示,上述数据银杏内酯组和NGF组之间的差异均无显著性意义(P>0.05),而他们与对照组之间的差异均有显著性意义(P<0.05或0.01)。结论:银杏内酯和NGF对体外培养的人胚肝细胞功能有促进作用。
【Abstract】 Objective: To study the effects of ginkgolide and NGF on the function of the cultured human embryonic hepatocytes in vitro. Methods: The hepatocytes were separated from human embryonic liver adopting modified non-perfusion method and cultured with DMEM medium. The cultured cells were divided into the ginkgolide group, NGF group and control group. At the 9th day of culturing, the glycogen staining was employed. The number and the grey volume of glycogen staining positive cells were counted and detected. The contents of albumine and urea in the medium were determined and analyzed by variance and Newman-Keuls test. Results: The number of glycogen staining positive cells was 74.63±12.76, 83.25±11.18 and 38.25±9.44 respectively in ginkgolide group, NGF group and control group. The grey volumes were 123.19±18.96 in ginkgolide group, 107.25±17.09 in NGF group and 164.91±21.65 in control group respectively. The contents of albumine(g/L) were 3.213±0.422, 3.450±0.481, 2.575±0.498, and the concentrations of urea(mmol/L) were 1.813±0.290, 2.013±0.344 and 1.363±0.220 in three groups respectively. The statistical analysis of above data showed that there were no significant differences between ginkgolide group and NGF group(P>0.05),but the ginkgolide group or NGF group were significant different from control group(P<0.01 or 0.05). Conclusions: Ginkgolide and NGF can promote the function of the primary cultured human embryonic hepatocytes in vitro.
