聚乙烯醇编织构建组织工程前交叉韧带的初步研究
Preliminary experimental study on the fabrication of tissue-engineered anterior cruciate ligament using the braided polyvinyl alcohol materials
【摘要】 [目的]探讨聚乙烯醇体外编织构建组织工程前交叉韧带支架材料的可行性。[方法]用聚乙烯醇纺丝纤维编织构建韧带支架材料,在电子拉力机上测试该支架材料的力学性质;用组织块和胶原酶消化培养法在体外分离培养人前交叉韧带细胞并对细胞的生长形态和分泌胶原蛋白的特征进行检测,细胞经体外扩增后种植于编织构建的聚乙烯醇韧带支架材料上观察。[结果]韧带支架材料的柔韧性强,拉力测试的负荷—拉伸曲线与韧带的拉伸曲线相似,其最大负荷、极限应力和弹性模量分别为52.61 N、14.96 MPa和202.08 MPa;体外分离培养的人前交叉韧带细胞呈典型的成纤维细胞特征,能在体外分泌Ⅰ、Ⅲ型胶原等细胞外基质;支架材料无细胞毒性,人前交叉韧带细胞可在支架材料上黏附、生长并分泌细胞外基质。[结论]支架材料具有一定的力学性能和优良的生物相容性,有望成为一种组织工程前交叉韧带支架材料。
【Abstract】 [Objective]To investigate the feasibility of the fabrication of tissue-engineered anterior cruciate ligament(ACL) in vitro using the braided polyvinyl alcohol(PVA) materials.[Method]The ligamentous scaffold material were made from the PVA fibers by braiding technique.The mechanical properties of the scaffold materials were measured using the electronic materials testing machine.Human ACL cells were isolated and cultured by tissue explant and collagenase digestion methods,the morphous characteristics of cells and the secretion of collagen protein were measured.In order to observe the biocompatibility between the material and cells,ACL cells were in vitro expanded and seeded on the scaffold materials.[Result]The flexibility of the scaffold material was powerful.The load-elongation curve of the mechanical testing was similar to that of ACL,the maximum load,the ultimate stress and the Young’s modulus of the scaffold materials were 52.61N,14.96 MPa and 202.08 MPa respectively.Human ACL cells displayed the representative characteristercs of the ligamentous fibroblasts,and synthesized extracellular matrix,such as type I and III collagen protein.The scaffold materials had no cytotoxicity.Human ACL cells adhered,grew and proliferated well both on the surface and in the holes of the scaffold materials.[Conclusion]The scaffold materials exhibited certain biomechanical properties and good biocompatibility,it may become a new biomaterial for constructing tissue-engineering ACL if the biomechanical properties can be improved.
