利用微波辅助提取测定姬松茸中麦角甾醇含量
Optimization of microwave-assisted extraction for determination of ergosterol in Agaricus brasiliensis.
【摘要】 建立一种简捷的微波辅助提取(MAE)方法以测定姬松茸菌丝体中麦角甾醇的含量.通过单因素实验和中心组合设计实验,对影响MAE提取率的因素进行筛选和优化.最优条件为乙醇和4mol·L-1KOH(4∶1比例混合)作为皂化提取剂,提取时间100s,微波功率520W,样品颗粒大小0.25mm(60~80目),皂化提取剂用量5mL(50mg样品),萃取剂用石油醚(60~90℃).在此条件下,MAE的提取率比传统的回流提取法和超声提取法提高约30%.和传统方法相比,MAE提取时间大幅缩短(从3h减少到只要100s),所需样品量和溶剂用量也大幅减少(分别从500mg和16mL减至50mg和5mL),并且重现性较高(RSD<5%).利用MAE法和紫外分光光度法相结合,姬松茸菌丝体中麦角甾醇的含量首次得到了测定,为(6.8±0.3)mg·g-1.
【Abstract】 A simple and rapid microwave-assisted extraction (MAE) procedure was developed and optimized for determination of ergosterol in Agaricus brasiliensis. The variables that can potentially affect the extraction efficiency were optimized using one-factor-at-a-time method and central composite design. Under the optimized operating conditions, i.e., alcohol to 4 mol·L -1 KOH (4∶1) as saponification solvent, extraction time 100 s, microwave power 520 W, particle size 0.25 mm (particles between 60 and 80 mesh), saponification solvent volume 5 mL (for 50 mg sample) and petroleum ether (60-90℃) as extractant, MAE showed significantly higher recoveries, about 30% higher, than those obtained by conventional reflux extraction and ultrasonic-assisted extraction. Furthermore, a drastic reduction of the extraction time (100 s versus 3 h), sample size (50 mg versus 500 mg) and solvent consumption (5 mL versus 16 mL), was achieved with a reproducibility (RSD<5%) compared with that provided by the conventional reflux extraction as a reference method. Using the new MAE method combined with UV-spectroscopy, the ergosterol content in mycelia of Agaricus brasiliensis was firstly determined as (6.8±0.3)mg·g -1 .
