针刺对脑出血大鼠神经生长因子基因表达的影响

【摘要】 目的:探讨头穴针刺对脑出血大鼠神经生长因子(NGF)基因表达的影响。方法:选取健康雄性Wistar大鼠160只,随机分为脑出血组、脑出血+针刺组(简称针刺组)、脑出血+脑复康组(简称脑复康组),每组又分为术后6 h、1 d、2 d、3 d、7 d五个时相点,每个时相点10只大鼠,另设空白对照组10只大鼠,制备脑出血大鼠模型,在光镜、电镜下观察各组大鼠脑组织形态学改变。运用原位杂交的检测方法,观察不同时相点头穴针刺治疗对NGF mRNA的表达。结果:针刺组大鼠NCF mRNA阳性细胞数及阳性细胞平均光密度值随不同时相点的变化而逐渐增加,至7 d时达高峰,与脑出血组比较有显著性差异(P<0.01),具有统计学意义。结论:头穴针刺治疗能够促进NGF的表达增强,对脑出血大鼠有神经保护作用。

【Abstract】 Objective:Approach the influence of scalp-acupuncture on the effect of the gene expression of the nerve growth factor (NGF) of the cerebral hemorrhage rat.Methods:Select 160 healthy male Wister rats,and the rats are divided into the cerebral hemorrhage set,cerebral hemorrhage to add the acupuncture set,cerebral hemorrhage to add the western medicine set randomly.And each set is divided into five phase point subgroups:6h,1d,2d,3d and 7d after intervening factors. Each phase point subgroup has 10 rats,preparing 10 rats to be used as another blank matched control set.To prepare the model of acute cere- bral hemorrhage with the methodd of injecting auto-arterial blood into the brain;to observe each rat’s brain tissue appearance and observe the changes by light microscope and electron microscope;to use the de- tection method of the hybridization in situ observing dissilarity phase point subgroup’s expression of NGF-mBNAin the rat’s brain tis- sue.Results:The population and the average optical d masculine cell was gradually increased with the variety of phase point,and reached the peak at 7d.Acupuncture groups was compared with model groups,the difference is extremely significant (P<0.01).It has the statistical significance.Conclusion:By scalp-acupuncture treatment to the cere- bral hemorrhage rat can promote the expression of NGF-mRNA in the rats’ brain tisue and have the neuroprotictive effect.
