青蒿琥酯治疗家犬卫氏并殖吸虫感染的效果观察

【摘要】 目的探讨青蒿琥酯(Art)对双倍体卫氏并殖吸虫病的治疗作用。方法1)卫氏并殖吸虫成虫用含不同浓度Art的RPMI 1640培养基体外培养后作电镜观察;2)实验犬常规感染卫氏并殖吸虫囊蚴,76 d后用青蒿琥酯50和80mg/kg灌胃治疗,103 d解剖犬,作常规病理检查,取出的虫体作电镜观察。结果1)5 mg/kg组体外培养24 h后,虫体萎缩,扫描电镜观察部分体棘消失、倒伏、絮状物附着;透射电镜显示皮层水肿、细胞器及分泌颗粒减少,卵黄腺萎缩,卵巢卵细胞胞浆空泡变等;2)两治疗犬体内虫体萎缩,厚壁囊肿数分别为28和35个,对照犬为16个,差异有显著性(P<0.05);透射电镜显示成虫变化与体外培养试验相似。结论青蒿琥酯对卫氏并殖吸虫有损伤作用。

【Abstract】 Objective To explore the therapeutic effect of artesunate(Art) on the experimental dogs infected with Paragonimus westermani.Methods 1) The adult worms of P.westermani were cultured in RPMI 1640 including Art in order to detect the toxicity of the drug;2) 76 days after the dogs were experimentally infected with the metacercaria of P.westermani,the intragastric administration was performed according to 50 mg/kg.The dogs were sacrificed for pathological examination with electron microscope after 103 days.Results 1) In vitro culture group,24 hours after 5 mg/kg Art were added,the worm body became depauperation,partly body disappear,lodging and cohered with floccules,the tegument became dropsy,the number of the cell organelles and the secretory granules decreased,the vitellarium depauperated,the cytoplasm of the ovum in female testis became vacuole;2) In the treated dogs group,the worm body depauperated,the number of the thickness wall cysts was more than that of the control group(P<0.05),the form changes of the adult worms were similar to in vitro. Conclusion Artesunate may damage the P.westermani.
