补肾中药对自然衰老雄性大鼠骨髓微环境中Smad1基因表达的影响
The Effect of Invigorating -kidney Herbs on Smad1 Gene Expression in Bone Marrow Microenvironment of Ageing Male Rat
【摘要】 目的:研究补肾中药对自然衰老雄性大鼠骨髓微环境中Smad1基因表达的影响。方法:以12月龄自然衰老雄性大鼠60只为模型,随机分为两组,每组30只。自鼠龄12月龄始,分别给予生理盐水、补肾方(密骨胶囊)灌胃,在给药8、16、24w末三个时间点,各时间点每组各取10只大鼠,取股骨骨髓,Trizol法抽提总RNA,RT-PCR技术半定量检测Smad1基因表达,采用天能GIS凝胶图像处理系统分析各组表达灰度值。结果:补肾方组大鼠骨髓微环境中Smad1基因表达明显优于生理盐水组。在8、16、24w末,补肾方组与生理盐水组的基因表达灰度值分别为0.61(95% CI=0.41,0.83)VS0.76(95% CI=0.56,0.98)、0.97(95% CI=0.81,1.13)VS0.61(95% CI=0.45,0.78)、1.75(95% CI=1.59,1.92)VS0.56(95% CI=0.40,0.73)。两组间差异有统计学意义(P<0.01)。结论:补肾中药(密骨胶囊)可以上调骨髓微环境中Smad1基因表达水平,可能是其促骨形成、修复骨损伤作用的分子机制之一。
【Abstract】 Objective:To study the effect of invigorating-kidney herbs on Smad1 gene expression in bone marrow microenvironment of ageing male rat.Methods:After being randomly divided to an intervention group and a controlled group with 60 twelve months old male rats, 30 in each group, aging male rats were intragastricly administrated with invigorating -kidney herbs and Sodium Chloride respectively .Three time points were set in week 8, 16 and 24 after intervention. Rat mRNAs in femoral bone marrow were extracted by Trizol in each group and at each time point. As followed by the instructions of Tanon GIS software, Smad1 gene expression was calculated .Results:At the time points of week 8, 16 and 24, Smad1 gene expression in the intervention group and the controlled groups were 0.61(95% CI=0.41,0.83)and 0.76(95% CI=0.56,0.98),0.97(95% CI=0.81,1.13) and 0.61(95% CI=0.45,0.78), and 1.75 (95% CI=1.59,1.92)and 0.56(95% CI=0.40,0.73), respectively. Compared to the controlled group, in each time point, significant increasing of Smad1 gene expression were documented in the invigorating -kidney group.(P<0.01).Conclusions:IInvigorating-kidney herbs seem to stimulate expression of Smad1 gene in bone marrow of ageing male rats, although the specific mechanism of promoting bone formation should be investigated further.
