正交试验法优选穿心莲有效成分的提取工艺

【摘要】 目的:优选AB-8型大孔树脂分离穿心莲有效成分(APNF)的工艺条件。方法:以柱效率和APNF对ADP诱导人体外血小板聚集的抑制作用为指标,采用正交试验法考察水提液浓度及上样量、吸附时间、乙醇质量分数及洗脱体积对APNF分离的影响。结果:优选分离条件为用1.5倍柱体积浓度为1g.mL-1的水提液上样,吸附1.5h,用13倍柱体积的55%乙醇洗脱。结论:以AB-8型大孔树脂分离的APNF对ADP诱导的人体外血小板聚集有较好的抑制作用,确定的吸附与洗脱条件简便可行。

【Abstract】 OBJECTIVE To optimize the extraction technique for the effective components(APNF) from Andrographis Paniculata with the macroporous resin of AB-8 by orthogonal test.METHODS The optimum extraction was investigated by column efficiency and the depressant effect of the ADP-induced platelet aggregation in vitro.The liquor concentration and volume,adsorbing time,alcohol concentration and volume were the three factors in the experiment.RESULTS The optimum extraction technique were as follows:1.5 times of column volume of 1 g·ml-1 liquor,adsorbing 1.5 hours,eluted by 13 times of column volume of 55% alcohol.CONCLUSION The APNF extracted by the macroporous resin of AB-8 can antagonize the ADP-induced platelet aggregation in vitro.The optimum extraction technique was convenient and feasible.
