甘草酸二铵对镉致小鼠急性肝肾毒性影响的实验研究

【摘要】 目的观察甘草酸二铵(DG)预处理对雄性普通昆明种小鼠急性染镉所致肝肾损伤的影响并初步探讨其机制。方法检测经低、中、高3个剂量DG预处理后急性染镉的雄性小鼠肝肾组织丙二醛(MDA)含量,谷胱甘肽过氧化物酶(GSH-Px),超氧化物歧化酶(SOD),过氧化氢酶(CAT)的活力,并观察病理组织学及细胞超微结构的变化。结果DG可剂量依赖性地增加染镉小鼠肝肾组织中SOD,GSH-Px及CAT的活性,并抑制其MDA的生成,受损的病理组织和细胞超微结构也明显减轻。结论DG预处理雄性普通昆明种小鼠可防护镉所致的急性肝肾损伤,其机制可能与DG能干预镉对小鼠肝肾的氧化损伤有关。

【Abstract】 OBJECTIVE To observe the protective effects of diammonium glycyrrhizinate(DG) on acute liver and kidney toxicity in male Kunming mice which were pretreated with DG,and attempt to explore its possible mechanisms. METHODS Before injecting CdCl2 to estabolish the cadmium-poisoning animal models,the mice were pretreated with DG.Then the following index were assayed,including the contents of malony diadehyde(MDA),the activities of glutathione peroxidase(GSH-Px),superoxide dismutase(SOD) and CAT in liver and kidney.The histopathology and ultra-structure of the liver and kidney also were observed.RESULTS The MDA contents in liver and kidney of mice pretreated with DG decreased,the activities of antioxidase(GSH-Px,SOD,CAT) increased with the dose-effect dependented relationship.The structure damage of liver and kidney was recovered obviously.CONCLUSION DG could affect the acute toxicity of cadmium in liver and kidney of mice.
