姜黄素对慢性应激大鼠不同脑区cAMP水平及腺苷酸环化酶活性的影响
Effects of Curcumin on cAMP Content and Adenylate Cyclase Activity in Brain Regions of Chronically Stressed Rats
【摘要】 目的探讨姜黄素对慢性应激大鼠不同脑区cAMP水平及腺苷酸环化酶(adenylatecyclase,AC)活性的影响。方法采用不同应激方式交替、持续应激20d,制成大鼠慢性应激模型,用开野实验观察大鼠的行为改变,用酶联免疫吸附实验(ELISA)、放射免疫分析法(RIA)测定额叶皮层和海马中的cAMP含量和AC活性的改变。结果慢性应激组大鼠与正常对照组相比,在开野实验中10min内穿越格数、竖立及探究次数均减少;ig给予不同剂量姜黄素(2.5,5,10mg.kg-1,每天1次,共21d)后,应激大鼠穿越格数、竖立及探究次数不同程度增加。慢性应激模型组大鼠较正常对照组额叶皮层、海马中cAMP含量和AC活性明显下降,姜黄素能明显拮抗慢性应激引起的cAMP水平和AC活性下降。结论姜黄素对慢性应激大鼠的自主活动和探究行为有明显的改善作用,这一作用很可能与增强慢性应激大鼠不同脑区AC-cAMP信号转导通路的功能有关。
【Abstract】 OBJECTIVE To explore the effects of curcumin on the cAMP content and adenylate cyclase activity in brain regions of chronically stressed rats.METHODS Rats were treated with different ways of chronic stress for 20 d,then the behavioral changes were determined by open field test. To verify whether the changes in the cAMP content and the adenylate cyclase activity occurred in frontal cortex and hippocampus of chronic stress groups versus control group of rats, ELISA and RIA experiments were performed respectively. RESULTS Chronic stress induced a decrease of locomotor activity in the open field test, which was significantly reversed by curcumin.Distinguished declines were obtained in the content of the cAMP and the adenylate cyclase activity in the stressed rats, while distinguished improvement in curcumin treated ones was obtained.CONCLUSION Curcumin significantly improves the number of ambulation and rearing of chronically stressed rats, which may be mediated by strengthening the function of AC-cAMP signal transduction cascade.
