运动对大鼠骨骼肌p38活性的影响

【摘要】 目的:探讨运动对大鼠骨骼肌p38磷酸化、蛋白和基因表达的影响。方法:SD大鼠随机分为对照组和运动组。运动组分为1h/d、1.5h/d组,共7周,运动结束后24h和48h取材。测定葡萄糖和胰岛素浓度。采用Westernblot法检测p38和phos-p38的表达,用RT-PCR法分析p38mRNA的表达。结果:与对照组比较,1h/d运动24h取材和1.5h/d运动24h与48h取材p38磷酸化水平明显升高;1.5h/d运动24h取材蛋白含量下降;p38mRNA升高发生于1h/d和1.5h/d运动24h取材组。结论:运动改善p38磷酸化、蛋白和基因表达。

【Abstract】 Objective:To study the effect of exercise on p38 phosphorylation and expressions of p38 protein and mRNA.Methods:SD rats were randomly divided into exercise group and control group.The rats in exercise group exercised for 1 or 1.5 hours per day for 7 weeks,and the blood sample was collected 24 and 48 hours after exercise.The serum levels of glucose and insulin were measured.The levels of p38 protein and phosphorylation were determined by Western blotting,and the level of p38 mRNA was determined by reverse transcription polymerase chain reaction.Results:Compared with control group,the level of p38 phosphorylation significantly increased 24 hours after exercise in rats that exercised for 1 hour per day and 24 and 48 hours after the exercise in those that exercised for 1.5 hours per day.The level of p38 protein 24 hours after exercise in rats that exercised for 1.5 hours per day was significantly lower than that in controls.The level of p38 mRNA significantly increased 24 hours after exercise in both rats that exercised for 1 hour per day and those that exercised for 1.5 hours per day.Conclusion:Exercise may improve p38 phosphorylation and the expressions of p38 protein and mRNA.
