肺癌与良性胸腔积液蛋白质双向电泳图谱差异分析

【摘要】 目的:建立肺癌与肺良性胸腔积液的双向电泳图谱,分析二者表达蛋白质的差异及特点。方法:利用固相pH梯度双向聚丙烯酰胺凝胶电泳法分离肺癌和肺良性胸腔积液中的总蛋白质。凝胶经银染色后用PDQUEST6.2.1电泳分析软件对胸水中的蛋白质谱进行分析,识别差异表达的蛋白质。结果:肺癌与肺良性胸腔积液的蛋白质组分布的基本框架很相似,并可发现二者之间有差异蛋白点;两种胸腔积液电泳图谱平均蛋白质点数分别为326±16和343±12,平均匹配点数为286±14和298±21,匹配率87.7%,86.9%.差异蛋白点82个,其中37个在肺癌胸腔积液中高表达,24个在肺癌胸腔积液中低表达。有17个点仅在良性胸腔积液表达,4个点仅在肺癌胸腔积液表达。结论:用双向电泳法得到了分辨率较高且重复率较好的肺癌与肺良性胸腔积液蛋白质组图谱,二者存在一些表达差异蛋白质。

【Abstract】 Objective:To establish 2-dimensional electrophoresis profiles with high resolution and reproducibility of the protein in pleural effusion caused by human lung cancer and benign diseases,and to analyze the difference in protein expression between these 2 types of pleural effusion. Methods:The total proteins in pleural effusion were separated by immobilized pH gradient 2-dimensional gel electrophoresis (2-DE). After silver staining,the differential protein expression was analyzed by using PDQuest 2-D analysis software. Results:The basic frames of proteomic distribution in these 2 types of pleural effusion were very similar,but each had different protein spots. In pleural effusion caused by lung cancer and benign diseases,the average protein spots were 326±16 and 343±12;the average matching spots were 286±14 and 298±21;the average matching rates were 87.7% and 86.9%. Eighty-two differential protein spots were identified. Among these 82 protein spots,37 spots were highly expressed and 24 poorly expressed in pleural effusion caused by lung cancer;17 protein spots were expressed only in the pleural effusion caused by benign diseases;4 protein spots were expressed only in the pleural effusion caused by lung cancer. Conclusion:The 2-dimensional electrophoresis profiles with high resolution and reproducibility of pleural effusion caused by lung cancer and benign diseases are established,and some differences in protein expression exist between these 2 types of pleural effusion.
