富集α-亚麻酸和维生素E的油菜花粉超临界CO2萃取条件优化
Optimization of Supercritical Carbon Dioxide Extraction Condition Enriched α-Linolenic Acid and Vitamin E from Brassica Campestris L.Pollen
【摘要】 目的:探明分离参数对油菜花粉超临界CO2萃取物分离比的影响,获得具有较高分离比,同时富集α-亚麻酸和维生素E的最佳分离条件。方法:选取分离釜1压力、分离釜1温度和分离釜2温度3个因素,利用正交表L9(34)进行正交试验,对试验结果进行直观分析及方差分析,得到合适的分离条件。利用GC-MS和HPLC分别测定萃取物中α-亚麻酸和VE的含量,并以此为参照确定最佳分离条件。结果:分离釜1压力对萃取物分离比有显著影响,最佳分离条件为:分离釜1压力7MPa,分离釜1温度60℃,分离釜2温度40℃。结论:调节分离参数,可以获得既具有较高分离比,又能富集α-亚麻酸和VE的最佳分离条件。
【Abstract】 Objective:To determine and to compare the influence of several separation parameters on separation rate,and to obtain an optimum separating condition with a higher separation rate and enrichment of a-linolenic acid and vitamin E. Methods:Three factors,pressure and temperature of first separator and temperature of second separator,were selected for orthogonal test with the orthogonal table L9(34). By using the range analysis and variance analysis,the appropriate separation conditions were obtained. The contents of a-linolenic acid and vitamin E determined by GC-MS and HPLC respectively were taken for reference to find out the best separating condition. Results:The pressure of first separator had significant influence on the separation rate,and the optimum parameters were 7 MPa and 60 ℃ in the first separator and 40 ℃ in the second separator. Conclusion:Through regulating the separation parameters,an optimized separating condition with a higher separation rate and enrichment of α-linolenic acid and vitamin E can be obtained.
