全基因组微阵列筛选候选雌激素非依赖型子宫内膜癌恶性标志物

【摘要】 目的明确非雌激素依赖型(Ⅱ型)和雌激素依赖型(Ⅰ型)子宫内膜癌在基因表达上的差异,找寻非雌激素依赖型子宫内膜癌的恶性标志物。方法利用全基因组Affymetrix基因芯片技术对2003年10月至2005年10月南方医科大学南方医院妇产科收治的3例雌激素受体(ER)和孕激素受体(PR)阳性及3例ER和PR阴性子宫内膜癌患者的子宫内膜癌标本进行研究,检测FOSB、NTRK2、JUN等基因在非雌激素依赖型中的表达情况,结合生物信息学分析,并采用半定量逆转录聚合酶链反应(RT-PCR)验证结果。结果在非雌激素依赖型子宫内膜癌中FOSB、NTRK2和JUN基因明显高表达。结论FOSB、NTRK2和JUN基因或许可以作为非雌激素依赖型子宫内膜癌的恶性标志物。

【Abstract】 Objective To further define differentially expressed genes between Type Ⅱ and Ⅰ of endometrial carcinoma and find malignant biomarkers associated with estrogen-nondependent endometrial cancer.Methods We examined six cases of endometriocarcinoma(including three ER and PR positive and three ER and PR negative cases) with Affymetrix genome-wide microarray,and found that three high expressed genes such as FOSB 、NTRK2 and JUN in estrogen-nondependent cases,combined with the analysis of bioinformatics,then we used the semiquantitative PR-PCR to verify the result.Results Three marked high expression genes including FOSB,NTRK2 and JUN in estrogen-nondependent endometrial cancer were involved in the roles of cell proliferation,cell differentiation and cell transformation.Conclusion That three genes mentioned above may be taken as malignant biomarkers of estrogen-nondependent endometrial cancer.
