已烯雌酚与人乳腺癌细胞WISP-2基因表达

【摘要】 目的探讨已烯雌酚(DES)对人乳腺癌细胞(MCF-7细胞株)WISP-2基因(wnt-1inducible signalingpathway protein-2)表达的影响及DES在乳癌发生中的作用机制。方法用四甲基偶氮噻唑蓝(MTT)法检测DES对人乳癌MCF-7细胞生长活性的影响;以免疫细胞化学法及逆转录-聚合酶链反应法检测DES对人乳癌MCF-7细胞WISP-2mRNA和WISP-2蛋白表达影响。结果DES使MCF-7细胞增殖率增加,其吸光度A570值明显上升,与对照组比较,P<0.05。以10nmol/L的DES分别作用细胞不同时间,A570值随着作用时间的延长而增加。DES可使MCF-7细胞WISP-2mRNA和WISP-2蛋白表达上升,WISP-2蛋白和mRNA的表达强度随药物浓度增加而升高,WISP-2mRNA和蛋白表达随DES作用时间的延长而增加,其效应类似于雌二醇。结论IDES能促进MCF-7细胞增殖,2DES能诱导MCF-7细胞的WISP-2基因表达增加,作用强度随剂量的增加而增加;随着作用时间的延长而上升。

【Abstract】 Objective To explore the effects of diethylstilbestrol on the expression of the WISP-2 gene in human breast cancer cell line MCF-7,and its mechanism.Methods To detect the proliferation by using MTT method.The expression of WISP-2 mRNA and its protein were measured using RT-PCR and immunohistochemistry approcah.The experiment set up 17-β estradiol as positive control.Results Diethylstilbestrol accelerated the MCF-7 cell proliferation,the value of A570 obviously raised as the time raised,compared with control(P<0.05);The expression on WISP-2 mRNA and its protein was increased,the difference had significance.And the effect was increased after exposed to diethylstilbestrol for more time.The expression of WISP-2 gene was increased,the difference had significanc.Its effects was similar to 17-β estradiol.Conclusion Diethylstilbestrol can accelerate the MCF-7 cell proliferation.Diethylstilbestrol can induce the expression of WISP-2 gene in the MCF-2 cell.Its effects was increased when concentration was increased with time growing.
