瑞芝清对人胰腺癌细胞COX-2和VEGF影响
Effects of nimesulide on expression of COX-2 and VEGF in human pancreatic carcinoma PANC-1 cells
【摘要】 目的观察瑞芝清对人胰腺癌PANC-1细胞环氧合酶-2(COX-2)和血管内皮生长因子(VEGF)表达的影响,探讨瑞芝清抑制肿瘤血管生成机制。方法对人胰腺癌PANC-1细胞进行培养,不同浓度瑞芝清处理细胞后,用免疫组织化学染色检测COX-2和VEGF蛋白表达的变化。逆转录-聚合酶链反应(RT-PCR)检测COX-2mRNA和VEGFmRNA表达的变化。结果100,200,400μmol/L瑞芝清作用胰腺癌PANC-1细胞48h后,COX-2和VEGF蛋白表达率与对照组比较明显降低(P<0.05),并且COX-2与VEGF之间具有良好的相关性(r=0.785,P<0.05);RT-PCR结果也显示,随着药物浓度的增大,COX-2mRNA和VEGFmRNA表达逐渐减弱。结论抑制COX-2表达及由此引起的VEGF表达降低,在非甾体类抗炎药抑制肿瘤血管生成机制中可能具有一定作用。
【Abstract】 Objective To investigate the effects of nimesulide on the expression of COX-2 and VEGF;and analyze the mechanism of nimesulide of inhibiting angiogenesis in pancreatic cancer.Methods PANC-1 cells were treated with various concentrations of nimesulide(selective for COX-2 inhibitor).The expressions rates of COX-2 and VEGF were detected by immuno histochemistry.The level of COX-2 mRNA and VEGF mRNA were measured by reverse transcriptase-polymerase chain reaction analysis(RT-PCR).Results The expressions rates of COX-2 and VEGF in 100,200,400 μmol/L group were significantly decreased more pancreatic cancer group than those in control group(P<0.05).The expressions between COX-2 and VEGF mentioned above showed fine correlation(r=0.785,P<0.05).RT-PCR revealed that the mRNA expression of COX-2 and VEGF were reduced with high concentration of nimesulide.Conclusion The inhibition of the expression of COX-2 and VEGF may play a certain role in the antitumor process of NSAIDs.
