【摘要】 目的通过评价芬太尼、司可林等药与戊巴比妥钠复合麻醉在急性心梗猪模型实验中的应用结果,建立一种较好的应用于大型动物实验的麻醉方案。方法28只猪被分成结扎组(AMIn=22)和假手术组(n=8)。AMI组动物开胸、结扎阻断心脏左冠状动脉的前降支(LAD 23、对角支处),假手术组开胸不结扎冠状动脉。手术动物使用氯胺酮(10 mg/kg)和阿托品(0.5 mg/只)肌肉注射进行诱导麻醉;开放静脉通道,术前缓注戊巴比妥钠(20 mg/kg)、芬太尼(0.008 mg/kg)进行基础麻醉;术中动物肌肉松弛采用司可林(5 mg/kg),并气管插管,联通呼吸机。结果使用该复合麻醉方案时,手术动物安静,手术中动物循环稳定,术后复苏迅速,手术实验质量提高。AMI组动物死亡率是20%;结扎后动物心电图S-T段明显提高;TTC染色:梗死区肉眼可见灰白色;光镜下:梗死区结缔组织纤维增生。结论该复合麻醉方案可以较好的应用于大型动物实验麻醉。更多还原

【Abstract】 Objective To evaluate the effect of combined anesthesia using Fentanyl、Suxamethonium chloride and Pentobarbotal sodium in pigs of acute myocardial infarction(AMI) and establish the optimal anesthesia in large animal experiments.Methods 28 young farm pigs were divided into operated(n=22) group and sham-operated(n=6) group.The left anterior descending(LAD) coronary artery distal to the second or third diagonal branch was ligated.Sham-operated animals underwent a similar procedure except that LAD was not lighted.Ketamine(10 mg/kg) and Atropine(0.5 mg/mL pig) were injected into operated pigs through IM to perform anesthetic inducement.Pentobarbotal sodium(20 mg/kg) and Fentanyl(0.008 mg/kg) were slowly injected by intravenous anesthesia before operation.The muscle of operated animals was relaxed by Suxamethonium chloride(5 mg/kg) during operation.The operated pigs were intubated,and mechanically ventilated.Results The pigs with Combined anesthesia had advantages as follow: stable circulatory function,quicker awakeness,and better quality of recovery.The mortality from AMI pigs was great reducing(20%).After LAD ligated,S-T elevation to single-phase curve appeared in the operated animals.The infarcted areas were offwhite by the TTC stain method.Light microscopy examination showed symmetrical fibrous connective tissue proliferation in the infracted area.Conclusion The method of combined anesthetic can be applied in large animal experiments satisfactorily.更多还原
