【摘要】 目的:采用合适的裂解液和沉淀方法提取乳腺癌组织蛋白质,获得高分辨率、重复性好的蛋白质双向电泳图谱。方法:通过不同全细胞蛋白质裂解液和不同沉淀方法提取组织全蛋白的效果比较,设计了不同的蛋白质样品制备的方法,并对2-DE的条件进行优化。结果:确定了适合乳腺癌实体瘤组织的裂解液配方(LS IV),PEG沉淀的蛋白分布较完整,沉淀效率相对较高。结论:本方法可以应用到乳腺癌组织的蛋白提取,也可对其它组织蛋白质样品的制备和双向电泳提供借鉴。更多还原

【Abstract】 Objective:To obtain the high resolution and better reproducibility two-dimensional gel electrophoresis(2-DE) of protein with breast cancer tissue extracted by suitable lysate and sediment methods.Methods: The effect of tissue total protein extracted by different total cell protein lysate and sediment methods were compared to design different method to prepare the protein sample and to optimize 2-DE.Results: The suitable lysate prescription(LSⅣ) for breast cancer tissue were set according the complete protein distribution of PEG sediment and the higher sediment rate.Conclusion: This method could be applied to extract the protein in breast cancer and prepare the other protein sample and 2-DE studies.更多还原
