青藤碱抑制TNF-α诱导人脐静脉内皮细胞VCAM-1表达

【摘要】 目的:研究青藤碱(SN)对肿瘤坏死因子-α(TNF-α)诱导脐静脉内皮细胞(HUVECs)VCAM-1表达的影响。方法:从新鲜脐带中分离培养HUVECs。用TNF-α诱导HUVECs表达VCAM-1,实验组加入不同浓度的SN(0.25、0.5和1.0 mol/L)或地塞米松(1.0×10-6mol/L)进行干预,培养12 h后收获细胞,用实时定量PCR检测VCAM-1 mRNA的表达,用流式细胞仪检测细胞表面VCAM-1表达。结果:TNF-α可诱导VCAM-1mRNA和VCAM-1的表达。进行药物干预后,各干预组相对VCAM-1 mRNA表达有不同程度下降(P<0.05)。SN(1.0 mol/L)和SN(0.5 mol/L)干预组细胞表面VCAM-1表达下降(P<0.05)。SN(0.25 mol/L)和地塞米松(1.0×10-6mol/L)干预组未显示对TNF-α诱导的VCAM-1表达有抑制作用。结论:SN可抑制TNF-α诱导的脐静脉内皮细胞VCAM-1的表达。

【Abstract】 AIM: To investigate whether sinomenine(SN) can decrease TNF-α-induced VCAM-1 expression in human umbilical vein endothelial cells(HUVECs).METHODS: HUVECs were isolated from freshly collected umbilical cords.Positive control samples were stimulated with TNF-α,omitting SN.Negative control samples were treated in the same way,omitting TNF-α and SN.Experiment samples were co-cultured with TNF-α and SN at different concentration(0.25,0.5,and 1.0 mol/L),or TNF-α and dexamethasone(Dex) at concentration of 1.0×10-6mol/L.Cells were harvested after cultivation with the drugs for 12 hours.VCAM-1 mRNA expression was detected by real-time quantitative PCR,and VCAM-1 expression was detected by flow cytometry(FCM).RESULTS: VCAM-1 mRNA and VCAM-1 were induced by TNF-α.Compared with the positives,the relative VCAM-1 mRNA expression decreased to varying degrees in the experiment groups(P<0.05),and SN at concentration of 0.5 mol/L and 1.0 mol/L inhibited expression of VCAM-1(P<0.05).SN at concentration of 1.0 mol/L decreased VCAM-1 expression by 28.8%(P<0.05),and SN at concentration of 0.5 mol/L reduced VCAM-1 expression by 21.68%(P<0.05).But SN at concentration of 0.25 mol/L and Dex at concentration of 1.0×10-6mol/L didn’t depress expression of VCAM-1.CONCLUSION: SN may inhibit TNF-α-induced VCAM-1 expression in HUVECs in vitro.
