Affymetrix基因芯片技术分析尖锐湿疣患者皮损局部免疫状态的研究
Analysis of Local Immunity in CA Lesions with DNA Microarray Technique
【摘要】 目的探讨尖锐湿疣(CA)患者局部免疫状态,分析局部Th1/Th2的变化。方法采用Affymetrix基因芯片技术分析CA皮损局部多个细胞因子和抗体变化,并用Real-timeQ-PCR验证部分结果。结果CA皮损局部Th1细胞因子IFN-γ,IL-12,TNF-α较正常包皮明显降低,Th2细胞因子IL-4,IL-10较正常包皮升高,IL-6降低,IL-2,IL-5,IL-13无明显变化;sIgA,IgA,IgG,IgM较正常包皮高。结论CA患者局部Th1/Th2细胞失衡,Th细胞向Th2极化,CA局部处于Th2优势应答状态。
【Abstract】 ObjectiveTo study the local immune and analysis the Th1/Th2 response in condyloma acuminatum(CA) lesion.MethodsTo analysis the levels of cytokine and antibody in CA local lesions with DNA microarray technique,and verify the results with Real-time Q-PCR.ResultsCompared with the normal control ,the intensities of Th1 (IFN-γ,IL-12,TNF-α) and Th2(IL-4,IL-10)type cytokines were lower and higher,respectively,and no change was found of IL-2,IL-5,IL-13.The transcript level of sIgA,IgA,IgG,IgM was increased.The data of Real-time Q-PCR was consistent with the microarrarys.ConclusionThe imbalance of Th1/Th2 response in CA lesions was verified,representing immune suppression of Th1 response and over activation of Th2 response.
