过敏性哮喘大鼠模型的建立及地塞米松对其的影响
Establishment of rat model of allergic asthma and the effect of desamethasone on it
【摘要】 目的:建立过敏性哮喘大鼠模型,并观察地塞米松对其的影响。方法:SD大鼠24只,随机分为对照组(A组)、模型组(B组)和地塞米松组(C组)。以卵蛋白致敏和激发建立大鼠支气管哮喘模型。C组大鼠激发前1h腹腔注射地塞米松0.5mg/kg。采用计数支气管肺泡灌洗液(BALF)中白细胞和嗜酸性粒细胞、及肺组织切片HE染色鉴定模型。结果:B组大鼠BALF中白细胞总数及嗜酸性粒细胞数明显多于A组和C组(P<0.01)。肺病理组织切片HE染色显示B组大鼠支气管收缩,部分支气管上皮脱落,支气管及小血管周围有大量嗜酸性粒细胞等炎细胞浸润,C组大鼠可见支气管周围有少许同样细胞浸润。A组大鼠未见明显异常。结论:成功建立过敏性支气管哮喘大鼠模型,地塞米松可减轻哮喘反应。
【Abstract】 Objective: To establish the rat model of allergic asthma,and study the effect of desamethasone on it. Methods: Twenty-four rats were divided randomly into control group (group A),asthma group (group B),and desamethasone (DXM) group (group C). Asthma rat model was established by sensitization and challenge with ovalbumin (OVA). One hour prior to OVA challenge,the rats in group C were intraperitoneally injected with DXM 0.5 mg/kg. The model was identified by the study of the total white cells and eosinophils counts in bronchoalveolar lavage fluid (BALF),The hematoxylin and eosin (HE) stain of lung histologic section of rats. Results: The number of total white cell,eosinophils in BALF in group B was significantly higher than that in group A and group C (P<0.01). Lung histopathologic slides of group B were bronchial constriction,part of epithelium shedding,characteristic of inflammatory cell such as eosinophils infiltration around the airways and blood vessels. But only a few of same cells were found infiltration around the airways and blood vessel in group C,and no abnormality was found in group A. Conclusion: The rat model of allergic asthma is established successfully,and desamethasone can relieve asthma attack.
