【摘要】 目的:考察珍珠菜总黄酮苷(ZTF)对HL-60细胞周期调控相关基因表达的影响。方法:通过逆转录-聚合酶链反应和琼脂糖凝胶电泳试验,考察不同浓度的ZTF对HL-60细胞p21、p27、cyclin B1和cyclin B2基因表达的影响。结果:ZTF对HL-60细胞p21及p27基因表达均有明显上调作用;在转录水平下调HL-60细胞cyclin B1的表达,而对cyclin B2表达影响不大。结论:ZTF可能通过上调p21及p27等CKI的表达,下调cyclin B1表达,对肿瘤细胞周期产生负性的调控作用,从而抑制肿瘤的增殖,最终导致肿瘤细胞凋亡。更多还原

【Abstract】 Objective: To investigate the influence of flavonoids of Lysimachia clethroides(ZTF) on cell cycle regulatory factors of HL-60. Methods: RT-PCR and agarose gel analysis were used to detect the different concentrations of ZTF on HL-60 gene expression p21,p27,cyclin B1 and cyclin B2.Results: ZTF could up regulate the expressions of p21 and p27 gene;down regulate the expression of cyclin B1 gene;but had little effect on cyclin B2.Conclusion: Through up regulating the expression p21 and p27 gene and down regulating the expression cyclin B1 gene,ZTF could negatively regulate cell cycle to inhabit the growth of tumor cell,and finally lead to apoptosis.更多还原
