砂海星酸性粘多糖的分离纯化
Isolation and Purification of Acid Mucopolysaccharide from Luidia quinaria von Martens
【摘要】 利用碱提法结合酶解法从砂海星中提取酸性粘多糖.粗品经三氯乙酸和二次酶解除蛋白后,用42%和80%的乙醇分级沉淀分别得到多糖Ⅰ和多糖Ⅱ,其总糖含量分别为71.6%、80.4%,蛋白质含量分别为1.52%和1.38%.多糖Ⅱ依次用离子交换层析(DEAE-52)和葡聚糖凝胶(G-50)进行分离、纯化,结果表明分子中带有比较均一的电荷,硫酸基离子鉴定反应为阳性,并且组分比较单一;用葡聚糖凝胶(G-100)对其分子量进行测定,其平均分子量约为12kD.
【Abstract】 Acidic mucopolysaccharides of Luidia quinaria von Martens are extracted by sodium-hydroxide method and enzyme-decomposition method.The gross products are deproteined by trichloroacetic acid and twice enzymic hydrolysis.The purified product solution is then precipitated in succession with ethanol(42% and 80%) to get acidic mucopolysaccharide fractions Ⅰand Ⅱ,of which the contents of total sugar are 71.6% and 80.4% while the contents of protein are 1.52% and 1.38%,respectively.Fraction Ⅱ is further separated and purified by ion-exchange chromatography(DEAE-52) and Sephadex(G-50) and further analyzed with Sephadex(G-100).The results show that fraction Ⅱ are composed of one components with sulfate.The average molecular weight of this acidic mucopolysaccharide is about 12 kD.
